Effects of SiO©ü, Crocidolite, and MMF on the Activity of Telomerase in Normal and Cancer Cell Lines

Abstract
In this study, the activity of telomerase was measured in normal, fibroblast, and lung cancer cell lines treated with silica, crocidolite, glass fiber, and ceramic fibers which have been well-known as the fibrogenic agents. Following results were obtained after cell lines co - cultured with 10§¶ /ml of various agents for 24 and 48 hours.
1. In normal cell lines, no telomerase acivity was observed in all conditions including control, even after being cultured for 24 and 48 hours.
2. In fibroblast cell lines, no telomerase activity was observed as shown in normal cell lines cultured for 24 and 48 hours.
3. In contrast to other cell lines, lung cancer lines showed strong telomerase activity in all conditions including control. However, no clear difference in telomarase activities was observed between control cells, and treated up to 48 hours.
Based on these results, we presumed that longer treatment is necessary so that fine particles introduced into nucleus and cause genetic alterations in vitro. Especially, further experiments concerning about relationship between telomerase activity and the amount or the incubation time with fine particles on the activities of telomerase in all three cell lines are needed.
